Wnt signaling is modulated by JARID2. a Activation of canonical Wnt signaling restores differentiation in JARID2 depleted cells. shJarid2 cells were treated with either 10 mM NaCl or 10 mM LiCl for 2 days in differentiation conditions before immunohistochemistry with myosin heavy chain (MHC) antibodies. Nuclei were stained with DAPI. Images were taken at × 400 magnification. Bars represent 100 μm. b, c Differentiation-specific gene expression is induced by Wnt activation. Expression of Tnni2 mRNA (b) and Mylpf mRNA (c) was quantitated by qRT-PCR. Fold change in expression of mRNA was calculated relative to undifferentiated scramble control. Data are plotted as means with S.E.M. and analyzed with one-way ANOVA followed by Tukey’s HSD post hoc test. Means not sharing same letter are statistically significant (p < 0.001). n ≥ 4
