Model for PRC1.6 function in cells with low or high Notch activity. L3MBTL2 and E2F6 binding facilitates establishing preferential environment for transcription factor RBPJ occupancy. In presence of a Notch signal and PRC1.6 complex, RBPJ is properly anchored and can efficiently transcribe (first lane). In the absence of either L3MBTL2 or E2F6 RBPJ chromatin binding is impaired and gene expression is downregulated (second lane). In cells with low Notch signaling activity, RBPJ binds to Notch target genes and represses transcription (third lane). In L3MBTL2 or E2F6-depleted cells, RBPJ occupancy is decreased (fourth lane)
