Predicted model by which EZH2 regulates DNA replication via dimethylation of PCNA. In normal cells, EZH2-mediated dimethylation of PCNA at K110 is required for the DNA replication performed by POLδ. Upon repression of the transcription or the methyltransferase activity of EZH2 or inhibition of the interaction between EZH2 and PCNA, stabilization of the PCNA trimer is perturbed, resulting in a defect in POLδ binding
